Allowed to settle at the tail of the field by James McDonald, the big bay loomed into contention on the turn and lengthened stride impressively to the line to win the 1100 metre dash by more than a length.
The John O'Shea trained colt was third on debut at Rosehill on October 31, so has plenty of room for improvement as he gets fitter and steps up in distance.
"He did a good job and gathered them in very quickly," said John O'Shea.
"He's a lovely colt with a great future. This will do him the world of good.
"When the races get longer he'll really come into his own."
A homebred for Darley, Telperion (images by Steve Hart) is the first foal of metropolitan winning Redoute's Choice mare Yavanna, a daughter of stakes-winner Wild Queen from the Crowning Jewel branch of the Denise's Joy family.
Yavanna has a yearling colt by Sepoy, a colt foal by Lonhro and was covered last spring by Reset.
Telperion is the 97th stakes-winner for Darley's former flagship shuttler Street Cry, who died in 2014 and is closing in on his century of stakes-winners.
